Calcium chloride stabilizes the bacterial cell membrane and cell viability and sodium thioglycollate acts as an anti-oxidant in a low oxidation-reduction potential environment. A minimum amount of agar is used to provide the consistency desired to protect the specimen in transit and methylene blue is used as a redox indicator to determine oxygen exposure. The sterile swab allows for easy collection of the sample and safe transport to the transport medium.
