Ponceau S, Sodium Salt is a dark brown to very dark reddish-brown powder that is commonly employed as a protein staining dye in molecular biology and biochemistry labs. The dye is particularly useful for the quick visualization of protein bands in membrane-based protein analysis techniques, as the staining is easily reversible. It is also routinely used for staining proteins on nitrocellulose, PVDF, and cellulose acetate membranes following immunoblotting and Western blotting. As the sodium salt of a diazo dye, Ponceau S allows clear visualization of transferred bands of protein, enabling researchers to verify transfer efficiency prior to further analytical procedures being performed. The membrane can then be re-used for further work, due to its reversible staining property.
