Description
Nutrient media are basic culture media used for maintaining microorganisms, cultivating fastidious organisms by enriching with serum or blood and are also used for purity checking prior to biochemical or serological testing. Nutrient Broth has the formula originally designed for use in the Standard Method for Examination of water and waste water. Nutrient Broth is one of the several non-selective media useful in routine cultivation of microorganisms. It can be used for the cultivation and enumeration of bacteria which are not particularly fastidious. Addition of different biological fluids such as horse or sheep blood, serum, egg yolk etc. makes it suitable for the cultivation of related fastidious organisms. The medium consists of Peptone, Beef extract and yeast extract that provide the necessary nitrogen compounds, carbon, vitamins and also some trace ingredients necessary for the growth of bacteria. Sodium chloride maintains the osmotic equilibrium of the medium.
Nutrient Broth finds wide application across different sample types including clinical materials such as urine and faeces, along with food, dairy, and water samples. Its simple formulation makes it suitable for routine laboratory workflows where consistent microbial growth is required without selective pressure. It is commonly used in laboratories for general cultivation purposes before proceeding to more specific identification methods. It is essential that samples be collected and handled correctly in order to obtain reliable results. Standard operating procedures should be followed according to the type of sample being tested and contaminated material should be sterilized after use to avoid contamination for laboratory safety.
Nutrient Broth is for professional use and appropriate precaution such as wearing of safety equipment and maintaining microbiological practices should always be observed during handling. Since this medium is intended for general use, certain fastidious organisms may need additional enrichment or a special type of media for best results. Therefore, further testing with specific microorganisms is recommended. When stored under appropriate conditions, this medium is stable and has no adverse effect on routine microbiological examinations.
