Description
Nigrosine (W/S) is a purplish-black crystalline dye commonly utilized in microbiology and microscopic staining procedures. It is an acidic dye which is soluble in water, and useful in negative staining methods. Nigrosine is commonly used as a substitute for India ink in the staining of spirochetes, bacteria, protozoa, and fungi. In negative staining procedures, the dye stains the background rather than the organism, allowing for better visualization of cell structures when viewed under the microscope. This unique feature of the dye makes it particularly interesting for microbiological observation and morphological analyses. The dye is a combination of synthetic black compounds derived from the heating of nitrobenzene, aniline, and hydrochloric acid in the presence of copper or iron. The consistent staining and ease of use made this dye ideal for research, teaching, and diagnostic laboratory applications.
