Haematoxylin is a yellowish-brown crystalline powder extensively used in histology and diagnostic staining procedures. It is an important microscopic examination because it provides cellular contrast, allowing clear distinction of the different elements of the cells and tissues. Hematoxylin (Delafield?s) serves as a stain in blood films for spirochaetes, protozoa, and other similar microscopic investigations. It is also a key component of the widely used Haematoxylin and Eosin staining technique, where acid-reacting parts of cells interact with alkaline dyes while alkaline cellular areas react with acid dyes. This staining combination allows high resolution examination of tissues in both pathology and research labs. The stain is also the best agent for demonstration of diagnostically important materials like amyloid, lipids, calcium, iron, melanin, hemosiderin, carbohydrates, and mucopolysaccharides.
