Description
GN Broth, Hajna is a selective enrichment medium designed to enhance the isolation of enteric Gram-negative bacteria with particular emphasis on Salmonella and Shigella from clinical and non-clinical samples. It is often utilized prior to plating on selective agar, especially when target organisms are expected to be in small numbers or mixed with competing flora.
The medium was supplied with the adequate nutrients by adding casein enzymatic hydrolysate and pancreatic digest of animal tissue. These components provide amino acids, nitrogen, vitamins and other factors of growth that are necessary for the restoration of bacteria. Dextrose is an energy source and sodium chloride is used for osmotic balance. High levels of mannitol promote the growth of mannitol fermenting Salmonella and Shigella, which facilitates their enrichment over some non-fermenting organisms such as Proteus species.
Selective agents such as sodium citrate and sodium deoxycholate inhibit the growth of gram-positive organisms and many coliforms, enabling better recovery of the target pathogens. Buffering salts help maintain a stable pH throughout incubation, ensuring consistent performance.
