Triton X-100? is a mild non-ionic detergent available as a colorless liquid. It consists of a large non-polar group which enables the molecule to associate with lipid membranes and hydrophobic protein regions. Upon addition to biological systems, it disrupts lipid bilayers, enhancing membrane permeability, without strongly denaturing proteins. This action helps release cellular components while maintaining the structure of many biomolecules. Due to this property, Triton X-100 is widely used in immunofluorescence staining for cell permeabilization. It is also used in immunoassay procedures to reduce non-specific binding. In biochemical experiments, detergents help with protein crystallization studies, affinity purification procedures, and buffer preparation for electrophoresis where a controlled solubilization of proteins is required.
