STERILE CHARCOAL SUPPLEMENT FOR LEGIONELLA AGAR

Sterile Charcoal Supplement for Legionella Agar is recommended for the cultivation and recovery of Legionella species. It is used as part of a supplemented Legionella Agar formulation to provide suitable conditions for the growth of these nutritionally demanding microorganisms. The supplement is combined with Legionella Agar Base and BCYE Growth Supplement during medium preparation.
The charcoal component supports the medium by helping reduce substances that may interfere with microbial growth. This contributes to a suitable environment for the recovery of Legionella during microbiological examination. The prepared medium is useful for laboratories performing isolation and cultivation of Legionella species from relevant samples.
For preparation, aseptically rehydrate the contents of one vial with 10 ml of sterile distilled water. Mix well to ensure the contents are evenly dispersed. Aseptically add the reconstituted supplement to 430 ml of sterile, molten, cooled Legionella Agar Base maintained at 45-50°C. Add one vial containing 50 ml of BCYE Growth Supplement. When no selective supplement is added, aseptically add 10 ml of sterile distilled water to bring the final volume to 500 ml. Mix thoroughly and pour into sterile Petri plates.
The final pH of the prepared medium should be 6.9 ± 0.2. Follow appropriate aseptic practices during preparation and handling.
