Description
Sabouraud Dextrose Agar is an acidic fungal culture medium employed for the isolation and cultivation of pathogenic as well as non-pathogenic yeasts, moulds and aciduric bacteria. It is especially appropriate for microbial limit testing of pharmaceutical products and it is recognized by the U.S. Pharmacopeia. USP General Chapters <61> and <62> describe the use of Agar for the enumeration of microbes and for the detection of Candida albicans in nonsterile pharmaceutical products.
The medium is buffered to pH 5.6 to promote fungal growth, especially dermatophytes and to inhibit bacterial contamination. The high dextrose content further promotes fungal growth and leads to selective conditions. Mycological peptone contains nitrogenous substances and other nutrients essential to requirements, while dextrose acts as the main source of energy. Agar provides the solidifying matrix required for plate-based cultivation.
The medium is suitable for recovery and total count of yeasts and moulds in environmental monitoring. For selective isolation of pathogenic fungi from samples that are heavily contaminated with bacteria or fungi, antibiotics can be included.
