PYR Reagent is intended for qualitative assessment of pyrrolidonyl arylamidase enzyme activity in β-hemolytic Streptococci. The test allows a quick biochemical differentiation of bacterial strains by their enzymatic activity, thus it is helpful for routine identification in the microbiology laboratory.
The solution of reagent was prepared by dissolving N,N-Dimethylaminocinnamaldehyde in hydrochloric acid and distilled water. The organism is initially streaked from the clinical specimen or suspected culture onto PYR Agar and incubated at 35-37°C for 18-24 hours for testing. Following incubation, one drop of PYR Reagent is added directly to the suspected colony. The reaction is then observed after approximately 2 minutes.
A red colour development indicates a PYR-positive reaction, demonstrating pyrrolidonyl arylamidase activity. No red colour indicates a negative reaction. The test can be used as part of a broader biochemical identification scheme and should be interpreted along with other relevant identification tests.
PYR Reagent is supplied in a 10 ml vial and should be stored in a sealed container at 2–8°C. Proper storage and handling help maintain reagent performance and support consistent results during routine microbiological testing.
