Description
Oxidase Reagent is used to perform the oxidase test for detecting cytochrome oxidase enzyme activity in microorganisms. This test is a key biochemical test to distinguish bacterial groups according to their respiratory enzyme systems.
The oxidase test was first used to identify Neisseria and later was found to be helpful in differentiating oxidase-positive organisms, including members of the Pseudomonadaceae, from oxidase-negative members of the Enterobacteriaceae. The reaction is a rapid preliminary assay that provides information on the metabolism of an isolated microorganism.
During testing, the suspected bacterial growth is brought into contact with the oxidase reagent using an appropriate testing method. Microorganisms possessing the target oxidase enzyme produce a characteristic colour reaction due to oxidation of the reagent. A rapid development of the expected dark colour indicates an oxidase-positive reaction, while the absence of the reaction indicates an oxidase-negative result.
The reagent has a blackish-purple coloured appearance and is intended for routine biochemical testing in microbiology laboratories. Results should be interpreted alongside colony characteristics and additional biochemical tests for complete identification.
