GRAM’S STAIN – KIT

Gram’s Stain Kit is used for differentiation of bacteria on the basis of their Gram reaction. Gram staining is a standard staining procedure that aids in the microscopic observation of bacterial cells, classifying them as Gram-positive or Gram-negative.
This kit employs a series of staining and decolourization steps. Crystal violet is first taken up by the bacterial cells and iodine acts as a mordant to form a dye complex within the cells. The decolourizing agent removes the primary stain from Gram-negative bacteria more readily than from Gram-positive bacteria which retain the crystal violet-iodine complex.
The decolourized cells are then counterstained with a contrasting color, so that the two groups of bacteria can be distinguished under the microscope. A 0.5% w/v Safranine or 0.1% basic fuchsin may be used as a counter stain depending upon the kit configuration.
The product has been tested and conforms to the specified quality standard. Preparation of smear, fixation, staining and decolorization are very essential for clear and reliable Gram differentiation.
