Description
C.L.E.D. Agar is used for differentiation and isolation of urinary pathogens based on lactose fermentation. C.L.E.D. (Cystine-Lactose-Electrolyte-Deficient) medium is prepared by the removal of sucrose and the addition of L-cystine for the stimulation of cystine-dependent dwarf colony coliforms. It is mainly used in urinary bacteriology. This medium has peptic digest of animal tissue, beef extract, and casein enzymatic hydrolysate to supply necessary growth nutrients. Lactose is an energy source that can be utilized by organisms through a fermentative process. Bromothymol blue acts as a pH indicator to distinguish lactose fermenters from lactose non-fermenters. Lactose-fermenting organisms will decrease the pH and alter the colour from green to yellow. L-cystine allows for the development of “dwarf colony” coliforms.
