Description
C.L.E.D. Agar W/ Bromo Thymol Blue is a selective and differential culture medium for isolation and differentiation of urinary pathogens based on lactose fermentation. It is especially effective in testing urine and permits the growth of a wide variety of urinary organisms while inhibiting the swarming of Proteus species.
Casein enzymatic hydrolysate and a peptic digest of animal tissue contain amino acids and other nitrogenous substances necessary for the growth of bacteria. The beef extract contains vitamins and minerals, and the lactose is the fermentable carbohydrate. Bromothymol blue is the pH indicator and turns medium green to yellow in the presence of acid production from fermentation of lactose, which distinguishes lactose-fermenting organisms from non-lactose-fermenters.
L-cystine is added as a growth factor for the cystine-dependent dwarf colony coliforms. The Proteus swarm is suppressed and other urinary pathogens may grow as discrete colonies by reduction of the electrolyte concentration through removal of the sodium chloride. Agar serves as the solidifying agent. The medium can be used with spread plate techniques or dip slides for the detection of bacteria in urine samples.
