5-Sulphosalicylic Acid 3% is used for the detection of proteins in urine samples by the turbidimetric method. The test provides a simple approach for identifying protein presence based on the precipitation produced when urine proteins react with sulphosalicylic acid.
The principle is based on protein denaturation by acid. Upon addition of sulphosalicylic acid to protein containing urine, protein solubility decreases and more of a solid like protein mass is formed which scatters the light resulting in a cloudiness in the solution. The degree of precipitation is related to the amount of protein present in the sample. This reaction can be employed for a qualitative or semi-quantitative determination of urinary proteins.
For reliable testing, the condition of the urine sample should be considered before performing the reaction. Highly turbid urine may mask a positive result, so the supernatant obtained from a properly centrifuged urine sample should be used. Highly alkaline urine may also affect the reaction and should be acidified to approximately pH 7.0 before testing.
5-Sulphosalicylic Acid 3% is suitable for routine laboratory analysis where detection of urinary protein is required. Proper specimen collection, handling, and established laboratory procedures should be followed to obtain dependable results.
