SOYA CASEIN DIGEST AGAR for isolation of various fastidious microorganisms as well as nonfastidious microorganisms with or without added blood. It is a good medium for isolation of anaerobes. This dehydrated medium is irradiated with dosage **25 kGy – 68kGy which is sufficient and adequate to eliminate sterility assurance level (SAL) of 10-6(5-8). Whereas, a minimum dose of 25 kGy is desired for microbial control or sterilization7. This medium is used for a multitude of purposes including maintenance of stock cultures, plate counting, and isolation of microorganisms from a variety of specimen types and as a base for media containing blood. Medium contains Pancreatic digest of casein and Soya peptone as sources of nitrogen, vitamins, amino acids and other necessary minerals. Sodium chloride is added for the osmotic balance of the media. Agar is a solidifying agent. The initial specimens should be inoculated onto another suitable medium and incubated for 18-24 hours in an aerobic atmosphere supplemented with carbon dioxide.
