For detection of microorganisms in blood.   BHI  Supplemented  w/  0.05%  is  used  for  detection  of  microorganisms,  including  aerobic  and  anaerobic bacteria. Brain  Heart  Infusion  Broth is  a  modification  of  the  original  formulation  of  Rosenow,  where  he added pieces of brain tissues to dextrose broth. Brain Heart Infusion Broth is also the preferred medium for   anaerobic   bacteria,   yeasts   and   moulds.The   media   is   especially   suited   for   the   cultivation   of staphylococci for the plasma coagulase test and for setting up blood cultures. Addition of ascites permits the  cultivation of gonococci. With the  addition of 10% defibrinated sheep  blood,  it is useful for  isolation and  cultivation  of Histoplasma  capsulatumand  other  fungi.  For  selective  isolation  of  fungi,  addition  of gentamicin and/ or chloramphenicol is recommended.BHI broth is also used for preparing the inoculum used in antimicrobial susceptibility tests.    
Beef  heart,  Brain  infusions  and  Peptone provides  organic  nitrogen,  carbon,  and  vitamins. Dextrose  is carbohydrate  source.  Low  concentration  of  dextrose  is  used  to  stimulate  early  growth.  Sodium  chloride maintains the osmotic environment. Disodium phosphate is buffering agent.    
BHI  Broth can  be  supplemented Sodium  polyanethol  sulfonate(SPS)which  isused  as  a  non-toxic anticoagulant  which  enables  bacterial  growth  and  prevents  the  action  of  natural  bacterial  inhibitors  of blood.

C.L.E.D. AGAR W/ Andrade Indicator (Cysteine Lactose Electrolyte Deficient Agar)

For isolation and differentiation of microorganisms based on lactose fermentation.   C.L.E.D  AGAR WITH  ANDRADE  INDICATOR is used for culturing of pathogens causing urinaryinfections. It improves colony detection and microorganism identification or it enhances the appearance of the colony and aids in the identification of microorganisms by the addition of the Andrade indicator. Andrade indicator is acid fuchsin in 1N sodium hydroxide. It is based on Lactose fermentation in whichend product is lactic acid which lowers the pH of the medium. Mixed culture is predominant in urine sample  and  test  such  as  coagulase,  oxidase,  catalase  and  agglutination  with  specific  sera,  can  be performed  on  primary  isolates  from  the  medium.  Beef  extract,  Peptone  and  Tryptone  provide  the nitrogen,  vitamins  and  amino  acids  in  CLED  Agar.  L-Cystine  is  added  as a  growth  supplement  for Cystine–dependent  coliforms.  Lactose  acts  as  a  fermentable  sugar.  Organisms  capable  of  fermenting lactose will lower the pH and change the color of the medium from green to yellow. Bromo thymol blue is used as a pH indicator. Agar is used as a solidifying agent.Inoculate and incubate at 35 ± 2°C for no longer than 24 hours, if lactose-fermenters predominate, the whole medium may turn pink, masking the presence of non-lactose fermenters.

For selective isolation of Vibrio cholerae & other enteropathogenic Vibrios causing food poisoning.      TCBS  AGAR  (VIBRIO  SELECTIVE  AGAR)  is  highly  selective  for  the  isolation  of Vibrio  choleraeand V.parahaemolyticusfrom stool specimens, and specified in standard methods for food testing.    
This highly selective medium meets the nutritional requirements ofVibriospeciesand allows competing with  intestinal  flora. Vibriospeciesis  able  to  grow  in  media  containing  increased  salt  concentrations. Vibrio speciesare natural inhabitants of water.      
Proteose peptoneand Yeast extract are the sole sources of carbon, nitrogen, vitamin B -complex, minerals and  amino  acids  in  the  medium.  Sodium  thiosulphate  serves  as  a  sulphur  source  and,  in  combination with   Ferric   citrate,   detects   hydrogen   sulphide   production.   Sucrose   is   included   as   a   fermentable carbohydrate  for  the  metabolism  of Vibrio.  Inhibition  of  gram-positive  bacteria  is  achived  by  the incorporation of Ox bile, which is a synthetic occurring substance and suppresses primarily Enterococci. Ox bilealso helps to inhibit gram -positive bacteria.       
The alkaline pH of the medium enhances the recovery of V. choleraebecause this organism is sensitive to acid  environments.  Thymol  blue  and  Bromothymol  blue  are  included  as  indicators  of  pH  changes. Sodium  citrate  and  Sodium  thiosulphate  are  the  selective agents,  providing  an  alkaline  pH  to  inhibit gram-positive  organism  and  suppress coliforms.  Agar  is  the  solidifying  agent.  TCBS  Agar deeps  (pour tubes)  are  provided  in  a  20  ml  fill  so  that  the  medium  may  be  liquefied  and  poured  into  a  petri  plates. This provides a convenient source of medium with a longer shelf -life than pre-poured plated media.

XLD  Agar  was  formulated  by  Taylor  for  the  isolation  and  differentiation  of  enteric  pathogens including Salmonella  Typhi from other  Salmonella species. XLD Agar has been recommended for the identification  of Enterobacteriaceae  and  for  the  microbiological  testing  of  foods,  water  and  dairy products. The media formulation does not allow the over growth of other organisms over Salmonella and Shigella.   

The  medium  contains  Yeast  extracts  as  source  of  vitamins  and  minerals.  Addition  of  Sodium deoxycholate acts as a selective agent which is inhibitory to Gram-positive bacteria. It suppresses the growth of other enteric pathogens and enhances the growth of only few enteric bacilli. The medium contains Xylose as a fermentable carbohydrate which is utilized by Salmonella species. The medium pH is changed due to fermentation of Xylose which is detected by indicator Phenol red, thus the colony colour turns red. The medium also contains Lactose and Sucrose as the source of fermentable sugar.  L-lysine  is  an  essential  amino  acid  source.  Lysine  is  added  to  differentiate Salmonella  Sp.  
Sodium  chloride  helps  maintaining  the  osmotic  balance  of  the  cells.  This  medium  also  allows differentiation of bacilli based on their ability to produce H2S; it contains Sodium thiosulphate and Ferric  ammonium  citrate  that  helps  visualizing  the  black  centered  colonies  on  production  of hydrogen sulphide in the medium. Agar is added as the solidifying agent.

TRANSPORT MEDIUM CARY-BLAIR W/O CHARCOAL  is  used  for  transportation  and  preservation of  bacteriological  samples,  without  significant  increase  in  growth.  Cary-Blair  Transport  Medium  is  a modification  of  Stuart’s  Medium.  It  is  a  chemically  defined,  non-nutritive,  low  oxidation/reduction potential  medium  to  maintain  the  viability  of  organisms  during  collection  and  transportation  of  the specimen(s), without allowing significant multiplication. Sodium chloride maintains the osmotic balance of  the  medium.  Disodium  phosphates  act  as  a  buffer system.  Agar  is  a  solidifying  agent.  Sodium thioglycollate and small amount of agar suppress oxidative changes and provide a reduced environment. An  alkaline  pH  of  the  medium  prevents  bacterial  destruction  caused  by  the  acid  produced.  Sterile  swab allows the easy absorption of specimen.

TRANSPORT  SWABS  W/  STUART  TRANSPORT  MEDIUM   is   used   for   transportation   and preservation of Neisseria species and other fastidious organisms. This medium was designed by Stuart for Gonococci. It is a chemically defined, non-nutrient medium suitable for transportation and preservation, without  allowing  significant  multiplication  of  microbes.  Calcium  chloride  salts  are  added  to  control  the permeability  of  the  bacterial  cell  wall  and  thus  prolong  their  survival.  Less  amount  of  Agar  is  added to make  medium  semi  solid.  Sodium  thioglycollate  suppresses  oxidative  changes  and  provides  a  reduced environment. Methylene blue acts as the redox indicator, the blue color indicates the presence of oxygen and it shows an adequate degree of anaerobiosis. Sterile swab allows the easy absorption of specimen.

MANNITOL SALT AGAR BASE

MANNITOL SALT AGAR (MSA) is a selective medium prepared according to the recommendations of Chapman for the isolation of presumptive pathogenic staphylococci.The coagulase-positive species of Staphylococcus i.e. Staphylococcus aureus is well-documented as a human opportunistic pathogen. S. aureus is also isolated from recreational water like swimming pools, and are thus indicators of health risk. S. aureus is relatively resistant to the effect of disinfectant like chlorine and sodium chloride. Staphylococci have the unique ability of growing on a high salt containing media. Isolation of coagulase-positive staphylococci on Phenol Red Mannitol Agar supplemented with 7.5% NaCl was studied by Chapman. The resulting Mannitol Salt Agar Base is recommended for the isolation of coagulase positive staphylococci from cosmetics, milk, food and other specimens.
